The equipment proposed by the PROTEOM'IC core facility

Preparation of the samples

  • A clean room is dedicated to sample preparation.
  • TECAN robot: the 8-channel "Tecan" digestion automaton is used to digest series of 96 samples from 1 or 2D polyacrylamide gels and then place them on a MALDI target for direct MS analysis or in microtiter plates for injection prerequisites in LC.
  • NanoHPLC chains: 4 Dionex Ultimate 3000 nanoHPLC chains "assist" mass spectrometers in electrospray ionization (ESI) mode. The facility is also equipped with Probot collectors for preparing samples for MALDI analysis.
  • Offgel Separation Apparatus: The Agilent Offgel apparatus separates peptides or proteins according to their isoelectric point and allows to recover them in solution after separation.

Mass spectrometry

  • timsTOF Pro: with funding from DIM Thérapie Génique 38%, GR-Ex 25%, Université Paris Cité 22%, IBiSA 12% and various 3%, the facility was equipped in June 2020 with 2 timsTOF Pro mass spectrometers. The timsTOF uses the PASEF (Parallel Accumulation Serial Fragmentation) acquisition method to provide extremely high speed and sensitivity to reach new depths in proteomics and phosphoproteomics with low sample amounts.
  • Orbitrap "Fusion": with 25% University, 25% Canceropôle and 50% EU funding, the facility acquired in 2015 a latest-generation Orbitrap-type spectrometer, allowing exceptional resolution and speed performance with the flexibility of 3 analyzers, 2 fragmentation modes and 2 detectors!
  • Orbitrap "Q-exactive": the facility was equipped in 2015 with a Q-exactive plus Thermo and two "Triversa nanomate" Advion sources (InCA, Ibisa, University funding) to meet the demand for analysis.
  • MALDI TOF/TOF: the facility has been equipped with an nLC-Probot-MALDI "4800 MALDI TOF/TOF" Absciex chain since 2007. This device is mainly dedicated to the analysis of purified peptides/proteins, for example following E2D separation

Electrophoresis

  • 1st dimension: 3 IPGphor (GE Healthcare) with Manifold for protein electrofocusing.
  • 2nd dimension: 1 Ettan-Dalt 12 device and 2 Ettan-Dalt 6 devices (GE Healthcare) for the migration of 12 and 6 SDS-PAGE gels respectively.
  • Scan & image analysis: 1 Typhoon FLA9500 fluorescence scanner (GE Healthcare) for digitizing 2D-DIGE gels and 5 DeCyder 7.0 licenses (GE Healthcare) for dedicated 2D-DIGE image analysis.
  • Spot picking: 1 Spot Picker robot (GE Healthcare) for spot picking of our E2D gels.
